Tex, this is my theory why I can not improve. I took Accutane I read that Isotretinoin affects Keratinocytes meaning the Suppression of lipid synthesis, downregulation of genes involved in keratinocyte proliferation, upregulation of genes involved in apoptosis Keratinocytes which are in the intetsines, stomach and mouth also modulate the immune system: they are a potent producers of anti-inflammatory mediators such as IL-10 and TGF-β. When activated, they can stimulate cutaneous inflammation and Langerhans cell activation via TNFα and IL-1β secretion. That is the cause of my LC
stage, increase of apoptotic cells, Ki67 that means Retinoid induced effluvium, and losing my hair It also affects Hair follicle keratinocytes which produce Premature Hair Folicule regression and change to catagen
Isotretinoin also affects Skeletal muscle producing Myalgia, muscle loss
ALSO AFFFECTS Hepatocytes BY Mitochondrial damage, cytochrome c release, induction of apoptosis, increased free fatty acids and liver triglycerides, THAT IS WHY I HAVE cholestasis.
Tex, I am so sad, if Accutane (isotretinoin) has changed my genes, I will never be able to get better right? Nor with diet, as it is most related to how Accutane messed my cells. Do you think may I have to take budesonide for the rest of my life? Even my hair never coming back

Hi Ingrid,

We have had other members in the past who had Accutane-induced MC. As far as I know, they have been able to achieve and maintain remission from their MC symptoms over the years. I don't believe any of them post anymore, so we probably can't ask them for an update to verify how they are doing. I don't remember who they were, anyway.

Isotretinoin has been known to cause MC for at least 13 or 14 years, I believe, maybe longer. But many doctors continued to prescribe it anyway. There have been many, many lawsuits against the manufacturers of Accutane.

As far as changing our genes goes, our genes are changing constantly, anyway. Many diseases cause gene changes, viruses can change genes, and many things in our environment can change our genes. Even stress can change genes. But most of those gene changes mean nothing unless they're triggered. And even if they're triggered, we can make changes in our lifestyle to overcome those changes so that they don't have a significant effect on our life. Worrying about gene changes will only cause more gene changes.

When I had my stroke, I'm sure I had many gene changes. Most of them were not much fun. But a few of them were actually beneficial. I've managed to overcome most of the bad changes, and I've appreciated the good changes. Life goes on.

At least that's my opinion.

Yes you can get better and get in remission. Almost everyone is able to get off of budesonide. A very tiny percentage
of folks are unable to get off of budesonide. Diet is very very key.

Youth is on your side. I see you are in your 30's. Those that are younger
heal faster. For those that are older....70's, 80's, 90's it takes much longer to heal.

Eating a lot of protein helped my hair and nails. Getting into remission helped my hair.

I was also on accutane when I was in high school. Might have been what started this journey. I took it for about 2 or 3 years.

In kinda remission now as long as I eat very carefully. I miss all the good things I used to eat.
